Digital Holographic Tomography for biological cellular imaging
Abstract
A new implementation of Digital Holographic Tomography (DHT) has been applied to biological cellular imaging in this thesis. The DHT technique provides quantitative information relating a cell's intracellular features to its variance in index of refraction or physical thickness. This can be used to monitor changes in a cell due to examples such as cancerous disease progression or the effects of drug delivery. The DHT technique has been integrated within the housing of a fluorescence microscope for increased robustness and user convenience. Three dimensional height and refractive index profiles of different blood cells have been generated. The results of these profiles validate and show promise in this novel technique.
